Most major airlines come into the Key West International Airport. Delta has a direct flight from Atlanta; US Air has a direct flight from Charlotte.
Also consider flying into Miami or Ft. Lauderdale and driving into the Keys.

Traveling by car:
Key West is approximately 3-and-one-half hours drive time from the Miami airport, and about 4 or 4-and-a-quarter hours drive time from the Ft. Lauderdale airport.
Marathon is approximately 2-and-one-half hours drive time from the Miami airport, and about 3 or 3-and-a-quarter hours drive time from the Ft. Lauderdale airport.
